We are considering this product as a wall finish, for above a wood-paneled wainscot. I see from the product specs that it is 1/2″ thickness. Should we first do a drywall or OSB substrate, or does the Homasote have enough rigidity to be directly fastened to studs?

I think you would find the Homasote waving at you after a few years. Also, it wouldn't have the fire prevention factor that rock does. Rock and tape first, then homasote.
