We are tiling 2500 sq ft using Ditra. Anyone have any thoughts pro/con of filling the Ditra holes in one step, letting it set and then thinset and tile? We would like a surface to do layout on.

never seen anyone do it but it, guess if the added time is worth it...
we normally snap lines with blue or black chalk for long runs, then use a sharpie to mark the ditra for a more permanent guide.
If you want to snap grid lines, try an ink line. Ink lines work great on Ditra.
If you're doing something more intricate, you can fill the Ditra, let it cure, snap lines, then tile later. I had a conversation with Schluter tech maybe 10 or 12 years ago and asked that question. They had no objections, no warranty issues, etc.

In the end we didn't pre-fill and I think it would have been a lot more work. We chalked our lines on the Ditra and hairsprayed them. It seems to be working well.
