I’m installing cedar clapboard siding over a vented air space. The cedar will be stained with a semi-transparent stain. Is back priming with the semi-transparent stain necessary?

It wouldn't hurt. However, it's much more important to be concerned about good moisture control than back primingd. As I'm not really sure what you mean by a vented air space (I assume you mean a crawl space under the home), I can't say with any certainty. But you can be sure of one thing: if you don't have good moisture control under the cedar, then back priming will only delay the inevitable for a few years. Some examples of moisture control are waterproofing concrete or concrete block, splines at the joints of your cedar, dripways and weep holes. Of course, there are others and it depends a lot on where you live.
